It's likely that your graphics card does not support OpenGL, which is necessary to play NS. What card do you have?

You should go to Options>Video and check what renderer you're using, change it to OpenGL if it's set to anything else.

BTW, is D3D preferable on any graphicscard?
Lt_Patch
Software Mode is an unsupported rendering method, which is only left in HL, for the sake of servers, and really old PCs. Like the error message says, you can't play NS in software mode.

Fix:
Start Natural Selection, go to Options, the Video.
Under Video Mode, make sure the dropdown shows either OpenGL or D3D.
Click OK, and wait for NS to reload in your chosen rendering mode.
Join a server as normal.
Enjoy Natural Selection.

Potential Issues With Above Fix:
1. If your video card is too old, then chances are it won't run in OpenGL or D3D mode.
2. If you're running the Microsoft drivers, then OpenGL, and D3D sometimes won't work

Workarounds For Potential Issues:
1. Buy a better graphics card, anything will do, as long as it's new.
2. Download, and install, the correct drivers for your graphics card. Reboot the PC, then try it again
